We are transported to the Soho neighborhood in London, England, in the year 1929. We see a frantic scene in a hospital, with the camera extremely shaky. Then, from a first-person perspective, we observe Tara Brooks, a waitress at Madame Vanity, who is about to give birth. Due to the heavy rain, there is no power in the hospital. As Tara faces severe complications during childbirth, a doctor is called. Dr. Deacon Frost appears, recreating his comic book pose (wearing a blue overcoat and with his arms crossed). He approaches Tara and recites the biblical passage from Revelation 11:08, which says: "Their dead bodies lie in the street of the great city, which spiritually is called Sodom and Egypt, where also their Lord was crucified." After leaving everyone confused, Deacon simply attacks Tara, drinking all her blood. This inadvertently transmits certain vampiric enzymes to her son when he is born. He is then attacked by police officers, and a killing spree begins in the hospital (the violence is extreme). The nurses arrive in time to save the newborn Eric, and one of them escapes with him. In the rain, she takes the baby to a young jazz musician known in the area as a former vampire hunter: Jamal Afari. She quickly explains the situation and leaves the baby and Tara’s diary with Jamal before fleeing. In an alley, Deacon, with a bloody face and licking his teeth, confronts the nurse who refuses to reveal the baby's location. He then violently kills her, while the camera pans up through the rain, capturing her agonizing screams. The scene cuts to the introduction of the Marvel Studios logo with the song "What a Wonderful World" by Louis Armstrong.

When the vampire refuses to cooperate, Blade tortures him. The vampire hands over a cell phone and a location of a bar where he would meet a mysterious woman. Blade then kills him. We see Blade return to England to meet his adoptive father and mentor, Jamal. Blade enters a room and examines a mural of vampire faces showing connections to Deacon. Inside, he finds Jamal preparing tea and listening to “It’s Been a Long, Long Time” by Louis Armstrong. Eric marks the faces of the dead vampires on the mural. He and Jamal begin to talk. Jamal asserts that Eric’s hunt was unproductive, that the target was a small fry, and that he should let go of his vengeful feelings. Eric responds that he is not seeking revenge but justice for his mother, using his skills to protect humans from the dark creatures that threaten society. His mission is to combat evil and eradicate vampires, partly as a form of justice for his mother and to ensure others do not suffer the same fate. After this father-son moment, Jamal says he has finished the research on the “Ebony Blade.” Eric thanks him but says Dane Whitman can wait (a reference to the post-credits scene from Eternals). The mural shows that Frost has used various disguises over the years and has contacts around the world, specifically an arms dealer in Madripoor named Abraham Whistler. They wonder if he might know something. Frost recites, “I am the lord of a vast domain and cannot allow anyone in without an invitation.” The camera then focuses on Bram Stoker’s book “Dracula.” In Madripoor, Blade arrives at the bar, takes out the two security guards, and enters for a drink while chatting with the bartender. Jamal says the mysterious woman might be nearby. A woman with brown hair appears at the bar to the sound of “Back to Black” by Amy Winehouse. She asks Eric, “I walk without legs, I cry without eyes. Wherever I go, darkness follows. What am I?” He replies, “A cloud.” She laughs and introduces herself as Rachel. They talk, and Rachel mentions Eric is older than she expected. She inquires about the Crystal of Nyx, but Eric claims not to know what it is. Rachel becomes enraged and tries to attack Eric, but he defends himself, calling her predictable. They start a hand-to-hand combat fight. Rachel proves to be very skilled in combat, but their fight attracts the attention of the vampires in the area, who corner them. They are forced to work together to fend off the vampires. Just when things seem lost, they are saved by a young hunter named Hannibal King but are captured and taken to Abraham Whistler. Abraham greets them, and Hannibal King asks if they are going to thank him. Rachel says he did nothing and only got in their way, while Blade remains silent. As the two argue, Blade tells them to stick to their plan. Abraham says Madripoor is crazy for bringing together three hunters. He is surprised to see Rachel Van Helsing, a descendant of Abraham Van Helsing, the famous vampire hunter. He expresses disappointment in seeing Hannibal King, a former henchman of his, and says he is honored to meet Blade, the famous vampire hunter. Rachel and Hannibal are surprised to see Blade, as he is a legend. Eric questions Abraham about Frost. Abraham says he is not an informant. When Eric asks why Abraham has been missing for 96 years, Abraham replies, “Sometimes, the eyes you don’t see are the ones that matter the most.” Jamal then cuts the lights, and Blade whispers, “Now.” He frees himself, Hannibal, and Rachel. The three join forces to fight the vampires. The fight is well-choreographed, with Hannibal using two katanas, Rachel using knives to attack and cut the throat of a vampire, all set to the song “Shining Star” by Earth, Wind & Fire. At one point, the scene becomes silent, and we see vampires running and shooting while Blade kills them amidst the flashes of gunfire.

The crystal’s surface also has an iridescent glow that appears to move like dancing shadows. It can turn darkness into a powerful weapon, allowing the manipulation of shadows and increasing the strength and agility of its user. The crystal was buried in a secret location known only to a handful of sorcerers and dark beings. The search for the Crystal of Nyx has become a legend among dark factions and artifact hunters. Deacon Frost, in his ambition to create a new vampire regime, seeks the crystal to amplify his own powers and subvert the established vampire order. The discovery of the Crystal of Nyx not only promises absolute power but also the chaos and destruction it could bring, as well as transforming the moon into a blood moon, amplifying its user to near-omnipotence and granting supernatural strength to vampires. However, a crucial part is missing to locate the stone: the blood of a half-blood. Eric thanks Wong and says he knows someone who can help. Jamal and Wong hug, with Jamal expressing how good it is to see Wong well and as the Sorcerer Supreme. Wong replies that it’s good to see Jamal as a family man protecting his son and allies. Rachel asks who they are going after next, and Eric responds: Dr. Karen Jenson, a hematologist who was his former romantic interest. They set out to find her. Dr. Karen Jenson is seen working in a hospital, performing a high-risk surgery. After some tense moments, she successfully saves the patient. As the fight reaches its dramatic climax, the music intensifies, reflecting the urgency of the situation, as Frost kidnaps Karen and escapes with her, leaving the group in total despair.

Karen asks why he is so obsessed with the Blood Moon and the Nyx Crystal. Frost reveals that he was betrayed by his own kind a long time ago and begins to tell his story, leading us into a black-and-white flashback: In the past, Deacon Frost had a wife named Eliza, a woman whose life was deeply intertwined with his. They met when Frost was still at the beginning of his journey as a vampire, before he became a leader within the vampire order. Eliza was a woman who believed in a peaceful coexistence between humans and vampires. They met in a café in old London, and soon Eliza's beauty and her thoughts on human-vampire coexistence made him fall in love with her. They shared the vision that such coexistence could be achieved, a vision Frost embraced early in their relationship. After several encounters and a ball, they fell deeply in love with each other. She stood by his side when he began to pursue a more radical path to transform vampire society, even though she did not agree with his methods. In the flashback, we see a dialogue between them where Eliza expresses her thoughts and opinions about their lives. We also see them with a third vampire (whose face is not revealed) going to an ancient catacomb where Eliza deciphers some riddles leading them to the Nyx Crystal. After Frost retrieves the crystal, he is betrayed by his hooded companion. During an internal crisis within the vampire order, Frost was betrayed by his own allies and friends. In an attempt to save the order or consolidate his power, these allies conspired against Frost, and his wife, Eliza, was killed in a orchestrated attack. Eric places his mother’s diary next to a photo of her and then adds a photo of himself with Jamal. He sends a message to Karen, asking if she’d like to grab coffee with him. Karen, currently at the hospital taking care of a patient, receives the message and agrees. The film ends with Eric and Karen enjoying coffee together, growing closer, accompanied by the song “Ain’t No Mountain High Enough” by Tammi Terrell & Marvin Gaye.

The fairy within it seeks your soul, and in failures we learn, not in the delirium of success.” The mysterious figure then declares, “That’s why I betrayed you and killed your wife back then.” Frost, barely conscious, whispers, “Dracula.” The figure removes their hood, revealing themselves as Dracula. “Your weakness and obsession led to your downfall, Frost,” Dracula says. He admits he has been watching every step until now and reveals that Blade has delayed his plans, but he knows what to do next. Frost asks what Dracula is doing there. Dracula responds, “I’m here to finish what I started years ago.” Dracula then coldly kills Deacon Frost, before slowly turning to the screen and the audience, opening a sinister smile.
